Denmark: Postdoctoral fellowship at The Novo Nordisk Foundation Center for Protein Research

Employment Conditions:

Employment will be in accordance with the provisions of the collective agreement between the Danish Government and AC (the Danish Confederation of Professional Associations). The position will be at the level of postdoctoral fellow. To the basic salary, a monthly contribution to a pension fund is added (17.1% of the salary), and a supplement could be negotiated, dependent on the candidate’s experiences and qualifications. Background:

The Novo Nordisk Foundation Center for Protein Research (CPR) has recently been established at the Faculty of Health Sciences, University of Copenhagen, to promote basic and applied discovery research on human proteins of medical relevance. The Center comprises a wide range of expertise and resources, from in silico target identification, proteomics, high throughput protein production and characterisation to chemical biology, disease mechanisms and protein therapeutics. To date, more than 70 scientists have been recruited (out of an expected ~150). We are now seeking a postdoctoral fellow to further strengthen collaborative efforts in the area of protein modifications and property profiling, within our activity area relating to protein therapeutics.


Project Description:

Chemical modification of therapeutic peptides and proteins is a key step towards modulating their pharmacological and therapeutic properties. The most common modifications are the attachment of polyethylene glycol (PEG) or acyl chains, and several such modified proteins are already in clinical use.

The chemical methods required to modify proteins have improved significantly during the last decade, now even allowing site- and/or region-specific modifications.

However, novel chemoselective methods are still of great interest to explore and apply to modifying proteins. These methods are currently being optimised in a recently initiated joint effort at the University of Copenhagen involving protein production and characterisation, protein chemistry and pharmaceutical profiling. The study is aimed at generating a comprehensive set of data from ~25 proteins, including both model systems and medically relevant targets. The proteins – both native and chemically modified – will undergo an extensive characterisation pipeline, involving both general physical as well as their pharmaceutical properties. To further strengthen these efforts, we are now seeking a highly skilled synthetic protein chemist to work on the development and implementation of novel chemical modification approaches, generally aiming at selective functionalisation of proteins.

Relationships: The post holder will jointly report to the Head of the Chemical Biology Unit at CPR (J. Berthelsen) and regarding chemical modifications strategies also to Prof. Knud J. Jensen (Faculty of Life Sciences). Work will in part be conducted in the laboratory of Prof. Jensen The post holder is expected to interact with staff at all levels, both internally and externally, regarding relevant research topics.

Job Description: To develop and implement chemical site specific modification strategies generally applicable to a wide range of proteins; preferably allowing reactions in buffered aqueous media at or near room temperature To develop and implement a modification approach for ‘chemical mutagenesis’ allowing incorporation of post translational modified residues To assist in the production and biophysical characterisation of proteins included in this study. To actively participate in strengthening interactions between the Center and the Faculties of Pharmaceutical and Life Sciences and other collaboration partners To actively participate in networks and collaborations with the international research community in general and the Danish research community in particular If required, participate in the supervision of students and technicians To carry out any other relevant duties which are reasonably associated with the post and could be required from time to time To demonstrate international reputation through scientific publications and presentations at international symposia.

Experience/skills required:

Essential criteria Excellence in synthetic bioorganic chemistry, including chemical modifications of proteins Proven track record in integrating organic chemistry with protein functionalisation or handling Experience in protein chemistry; e.g. purification and characterisation of recombinant proteins Good scientific publication record Excellent communication skills, both oral and written Integrity, enthusiasm, motivation, flexibility, confidence and outstanding collaboration skills.

Desirable criteria Experience with protein engineering, including site-directed and deletion mutations as well as fusion protein strategies Interest in devising novel approaches in selective functionalisation of proteins.

Qualifications required:

Highly motivated and ambitious candidates are encouraged to apply. A PhD in protein chemistry, biochemistry or synthetic chemistry is required. Solid experience and an excellent track record gained in leading laboratories in the research field will be considered a distinct advantage.
