This project is based on the idea that toxic secondary bile acids are central to the onset and progression of colorectal cancer. We have made two new observations in the field: i) we identified a variant of the cytoplasmic Ileal bile-acid Binding Protein (IBABP) that is essential for the survival of colon tumor cells (Fang et al., Cancer Research 2007), and is a likely drug target, ii) we have also established that IBABP is necessary for the transcriptional activation of FXR, providing a unique mechanistic insight into the biological connection between bile acids and transcription. Prospective fellows should have direct training in one or more the following areas: tumor biology of colorectal cancer, cell biology of colorectal cancer, bile acid biochemistry and enzymology, synthesis of bile acid analogs, or animal models of colorectal cancer.
